动脉粥样硬化兔不同金属基质蛋白酶动态变化的探讨

【摘 要】目的探讨不同基质金属蛋白酶在动脉硬化兔模型的动态变化。方法 高脂喂食加球囊拉伤股动脉建立动脉硬化兔模型,分别于拉伤后1、7、14、21、28 d采血,ELISA法检测MMP-1、2、8、9,TIMP-1,绘制MMPs动态变化曲线,并取血管行病理检查。结果 MMP-1、9,TIMP-1在拉伤后1 d均达到峰值,延续至拉伤后7 d;三者衍变曲线相似,且有相关性;MMP-2、8在各采血点无差异。结论 MMP-1、9,TIMP-1的动态变化规律可为研究动脉硬化不稳定斑块提供依据。

【Abstract】 Objective To search the dynamic changes of the matrix metalloproteinases (MMPs) and tissue inhibitors of metalloproteinases-1 (TIMP-1) of the atherosclerosis rabbit model.Methods Atherosclerosis models of rabbits were established through high lipid diet and injured femoral arteries by dilated balloon,and on day 1,7,14,21,28 after injured,MMP1,2,8,9 and TIMP-1 were detected with ELISA assay,then the curve reflecting the dynamic changes was draw,and the histopathology data was provided as well.Results The curve of dynamic changes MMP-1,9,and TIMP-1 were similar,the peaks were showed at 1 day after injured,and last for 7 days;there are relative among them;no obvious changes were showed in MMP-2,8.Conclusion The characteristic of dynamic changes of MMP-1,9 and TIMP-1 could provide evidence for search the vulnerable plaque.
