RP—HPLC法测定冻干粉针中DHEA的含量

[摘 要] 目的：建立反相高效液相色谱法测定冻干粉针中DHEA含量。方法：采用Diamonsil C18 （250 mm×4.6 mm，5 μm）色谱柱，流动相为乙腈-水（55：45），流速1.0 mL > min- 1；检测波长206 nm；进样量20 μL ；柱温：室温。结果：在此色谱条件下DHEA与有关物质能达到较好分离，在0.01 mg > mL- 1～
0.7 mg > mL- 1浓度范围内与峰面积呈良好线性关系， r=0.9998；方法重复性RSD为0.42%；加样平均回收率（n=6）为98.91%。结论：该方法准确可靠，灵敏度高，专属性好，可用于测定DHEA冻干粉针含量。

[Abstract] Objective： To establish an RP-HPLC method for determination of the content of DHEA. Methods：Diamonsil C18 （250 mm× 4.6 mm，5 μm） column was adopted ； the mobile phase consisted of a mixture of acetonitrile - water （55：45） at the flow rate was 1.0 mL>min- 1 ；The ultraviolet detection wavelength was set at 206 nm； Loaded volume was 20 μL； the column temperature was room temperature. Results： DHEA and its related substances could be separated well. The linearity of DHEA was well correlated （ r=0.9998） within the range of 0.01 mg > mL- 1～0.7 mg > mL- 1. The method precision RSD was 0.42%. The average recoveries （n=6） was 98.91%. Conclusion： The method appeared to be accurate， reliable， sensitive， specific and can be used for the determination of the content of freeze-dried powder injection DHEA .
