稻瘟病拮抗放线菌CZ133的筛选、鉴定及抑菌活性分析

摘 要:采用平板对峙法,从湖南水稻根际土壤样品中筛选对水稻稻瘟病具有抑菌活性的拮抗放线菌,通过菌落和菌株形态、16S rDNA和gyrB基因序列对拮抗放线菌进行鉴定,并分析了其发酵滤液对稻瘟病病原菌菌丝生长和孢子萌发的影响。结果显示:分离纯化得到了1株对稻瘟病具有显著抑菌活性的拮抗放线菌CZ133,其对稻瘟病病原菌的平均抑制率为59.47%,经鉴定属于链霉菌属;该菌株发酵滤液能显著抑制稻瘟病病原菌菌丝的生长,最高抑制率达100%,2.5%的发酵滤液能明显导致稻瘟病病原菌菌丝膨大或异常扭曲、甚至自溶,5%~50%的发酵滤液能明显抑制孢子的萌发,最高孢子萌发抑制率达74.27%。

Screening and Identification of Antagonistic Actinomycete CZ133 Against Rice Blast and Analysis of Its Antimicrobial Activity

Abstract: Antagonistic actinomycetes with antimicrobial activity against rice blast were screened from rhizosphere soil samples of Hunan rice by plate confrontation method. Through the morphology observation of colonies and strains, the antagonistic actinomycetes were identified by 16S rDNA and gyrB gene sequences, and the effects of fermentation filtrate on mycelial growth and spore germination of rice blast pathogen were analyzed. The results showed that an antagonistic actinomycete CZ133 was isolated and purified, and its average inhibition rate to the pathogen of rice blast was 59.47%. It was identified as Streptomyces. The fermentation filtrate of the strain could significantly inhibit the growth of the pathogen, and the highest inhibition rate was 100%. 2.5% fermentation filtrate could obviously cause mycelium enlargement or abnor...
