加味通幽汤对缺氧条件下食管癌细胞血管生成拟态和增殖的影响
【摘 要】 目的：观察加味通幽汤对缺氧导致的食管癌细胞血管生成拟态（VM）形态学和细胞增殖的影响。方法：缺氧环境培养Eca109细胞，镜下观察细胞VM，以加味通幽汤含药大鼠血清干预细胞，观察其对VM形态的影响，CCK8法检测方药对细胞增殖的影响。结果：常氧对照组细胞形态成团状且无规律；含药血清常氧组细胞分散，出现凋亡；缺氧对照组细胞相互连接呈管状；含药血清治疗组细胞成团块状并且出现凋亡。CCK8细胞增殖实验显示，含药血清治疗组明显低于其他3组（P=0.000）。结论：加味通幽汤可改善食管癌细胞VM形态变化，并抑制缺氧环境下的细胞增殖。

Abstract：Objective To observe effectof Modified Tongyou Decoction （MTD） onvasculogenic mimicry （VM） morphology and cell proliferation of esophageal squamous carcinoma cellunder hypoxic conditions.Methods Eca109 cells were cultured in hypoxia environment， vasculogenic mimicry was observed through microscope. Rats’ serum with MTDwas taken into culture medium， VMmorphology was observed and cell proliferation was determined with CCK8 assay.Results Cells in the controlled group shape of cells were colony and irregular； cells were scatteredin the medicated-serumcontrolled group， apoptosis emerging； however， cells were connected with each other in hypoxia group cell with fusion of tubulous structure； cells in the medicated-serum treatment group were mass clumps and apoptosis also occurred. Proliferation ratio in the medicated serum treatment group was significantly lower than that in the other 3 groups（P=0.000）.Conclusion Under hypoxia conditions，MTD shows significant effect on VM andproliferationof Eca109 cells .
