基于可开启微流控芯片的循环肿瘤细胞捕获及单个细胞的提取

摘要:介绍了一种可开启微流控芯片。基于细胞的物理尺寸特性该芯片可对外周血循环肿瘤细胞(circulatetumorcells,CTCs)进行分选富集及单个细胞的提取。可开启微流控芯片的结构简单,简化了制作流程。该文以前列腺癌细胞PC3为目标靶细胞,结合显微切割原理,实现了对该细胞分选富集、转印及单个细胞的提取,从而提高了CTCs检测的纯度。本文主要研究了可开启细胞捕获芯片的制作及单个细胞提取的方法。结果表明该芯片可以高效捕获到循环肿瘤细胞并且能从捕获到的细胞中任意提取单个目标细胞,从而为后续探讨肿瘤细胞的生物学机制和转移机制提供了很好的基础与平台。

Circulating Tumor Cell Capture and Single Cell Extraction Based on an Openable Microfluidic Chips

Abstract: An openable microfluidic chip is introduced,which is based on the physical size of the cells, for eiching the peripheral blood circulating cells (CTCs) and extract singlecell. The chip has simple structure and simplifies the production process. In this study, the prostate cancer cellsis used as the target cell, combined with the principle of micro-cutting, achieved the cell sorting eichment, transfer and single cell extraction, thereby enhancing the CTCs detection purity. In this paper, we mainly study the fabrication of cell-capture chip and the method of single cell extraction. The results showed that the chip could efficiently capture the circulating tumor cells and could extract a single target cell from the captured cells, thus providing a good foundation...
