CleistanthinA对黑素瘤细胞迁移及β—catenin蛋白的影响
【摘 要】
目的:研究Cleistanthin A(CA)對黑素瘤细胞迁移及β-catenin蛋白的影响。方法:通过划痕实验观察CA对A375细胞迁移活性的影响;通过Western blot实验观察CA对A375细胞中β-catenin蛋白的影响。结果:划痕实验显示:CA对A375细胞的迁移有抑制作用,且有剂量依赖性,且0.1μM这个浓度接近半数迁移抑制率;结合我们前期的实验得知:CA对A375细胞的生长活性有抑制作用,且有剂量依赖性,但低浓度的CA对A375细胞的活性抑制作用与阴性对照组相比虽有统计学意义却不显著[1],因此选用0.03μM、0.1μM、0.3μM较低浓度组CA进行后面的实验,以降低细胞凋亡对迁移的影响。Western blot实验显示:CA对A375细胞中β-catenin蛋白表达有抑制作用,且有剂量依赖性,0.3μM浓度时与阴性对照组相比有统计学意义。结论 CA可抑制A375细胞的生长活性及迁移且可下调与生长及迁移密切相关的β-catenin蛋白的表达。

Abstract Objective:to investigate the effect of Cleistanthin A(CA)treatment on melanoma cell migration and β-catenin. Methods The inhibitory activities of CA on cultured A375 cell migration were detected by Wound-healing assay;Western blot were used to detect the activity of β-catenin. Results Wound-healing assay shows:CA has obvious inhibitory effect to A375 cell migration activity, and in a dose dependent manner, and the concentration of 0.1μM is near the median inhibition rate. Combined with our previous experiments, we learned that: Cell viability is affected significantly by CA. It appears in a dosage manner. But the viability of A375 is not obviously affected by CA in low concentration, So the concentrations of 0....
