CFA致炎大鼠脊髓腰膨大组织内miRNA—133b—3p与P2X4受体表达的实验观察

【摘要】 目的:观察CFA致炎大鼠脊髓腰膨大组织内miRNA-133b-3p与P2X4受体表达的相关关系。方法:大鼠足底注射CFA后,0 h、2 h、1 d、3 d检测大鼠机械痛和热痛的痛阈值变化,且在这四个时间点取脊髓腰膨大组织,提取总RNA和蛋白质,检测miRNA-133b-3p表达量的变化,同时检测P2X4受体mRNA和蛋白的表达量的变化。结果:与对照组相比,大鼠脚掌注射CFA后,机械痛和热痛痛阈均明显下降(P<0.05),在2 h时痛阈最低,1 d和3 d时较2 h略有回升;腰膨大部位miRNA-133b-3p表达量在2 h时没有变化,1 d和3 d开始呈现逐渐下降的趋势(P<0.05),四个时间点P2X4受体mRNA表达量比较差异均无统计学意义(P>0.05),但P2X4受体蛋白在3 d时表达量显著升高(P<0.05)。结论:CFA致炎大鼠脊髓腰膨大组织miRNA-133b-3p表达下调,同时P2X4受体表达上调,两者可能存在负向调控关系。

【Abstract】 Objective: To observe the expression pattern of miRNA-133b-3p and P2X4R in lumbar spinal cord of complete Freund’s adjuvant(CFA)-induced inflammatory pain rats.Method:CFA was intraplantar injected to the rats.Then we measured the threshold to mechanical stimuli and noxious thermal stimuli at 0 h,2 h,1 d and 3 d after CFA injection.After the behavior observation,the lumbar spinal cord were isolated from the rats,and the total RNA and protein were extracted for the detection of miRNA-133b-3p,P2X4R mRNA and P2X4R protein.Result:Compared with the control group,the threshold of rats to mechanical stimuli and noxious thermal stimuli decreased and peaked at 2 h after CF...
